I agree the spot is small and no one but you will notice. When DH was working he would often get ink stains on his poly-cotton shirts. I would make a paste of dishwasher powder and rub it on the spot then wash it and the spot would come out. Lately, I use the gel dishwasher stuff, but it seems to work as well. Never tried hair spray.

It's really almost invisible & I think it would be fine to leave it as is. If you're pretty sure it's ink, the best way to get out ink is with hairspray. Just spray a little bit on the dry quilt in that area & gently rub (either rub against the quilt itself or rub with a soft white cloth). When all the ink is gone, you can wash the quilt -- or just spot wash the area with a teeny bit of laundry soap & cold water to remove the sticky hairspray. Works every time & I've yet to see it damage any type of cotton fabric.

No need to worry in my opinion, not anything to bother with.
But just an FYI - hand sanitizer also works to remove ink marks. Took several red ink marks out of a white blouse with no problem for me. Little dab on old, clean toothbrush, apply to area, rinse, gone!
