Do I need to have all of my quilt marked before I sandwich and begin quilting? I have only marked 1 pattern that I know I want. Just not sure what to do with the rest yet. Do i wait to quilt or what do I do now? Some will be by hand and some by machine. which do I do first?

What part of the quilt will you be machine quilting? If you are going to stitch in the ditch around large blocks and then hand quilt inside the blocks, I would do the STD first. If you're planning to do a large hand quilted motif in the center, I would do that first and then machine quilt out from it.
As for marking, some people mark their whole top first and then quilt. Some like to mark a section at a time. It all depends on what method of marking you are using. Chalk and a pounce is usually done a section at a time? A water soluable quilt marker is done all at once usually.

Tartan had great points.
You could also mark the designs for machine quilting on something like Golden Threads paper and then pin that to the machine quilting area.
You can also baste your quilt with washaway thread. Then quilt as you desire [keeping it dry of course.] In which case i would do the machine part first to help anchor it all while I do the hand quilting.
