neighbour gave me bag square fabric fron 5x5 down to 3x3 my question is can i make a quilt from odd squares ?

what filling do i use

bought my 5x5 steel template today and a smaller one for 1/4 seam
got my steel ruler and brand new cutter (hubby treated me )
got a chalk pen new needles thread
squares pre cut got...

as u now im starting my first quilt im wondering can i do my square put wadding on and back it on each square before i sew all togher or must i do all top squares then full wadding full...
